Treponema pallidum is the causative agent of syphilis and has never been successfully cultured in vitro, despite extensive attempts. It requires living hosts (humans or animals) to grow and cannot be grown on artificial media. In contrast, Leptospira, Bordetella, and Staphylococcus are all culturable using standard laboratory media. Leptospira grows on selective media, Bordetella on blood agar, and Staphylococcus on nutrient agar. The inability to culture T. pallidum is a key limitation in studying its biology and pathogenesis.
