In alkaptonuria, large amounts of homogentisic acid are formed which are excreted in urine or deposited in tissues. Nitisinone is a synthetic reversible inhibitor of 4-hydroxyphenylpyruvate dioxygenase. Nitisinone efficiently reduces homogentisic acid production in alkaptonuria.
